A spoonful of soup would consist of the soup being IN the spoon (or the concave portion that holds it) because if it were ON the spoon, that would probably mean that you dropped your spoon in the soupbowl and just fished it out and it's got soup all over it and now you have to get a clean spoon from the silverware drawer.
